Route Markers[]
The race begins at Castaic Lake in Castaic, California, United States.
Teams must run through a mud run obstacle course by the lakeshore. Once both members have finished the course, they may retrieve a ring that would contain tickets for their first destination: Tokyo, Japan. The first eight teams would receive tickets for the first flight, EVA Air, while the last three teams would receive tickets for a China Airlines flight scheduled to depart 45 minutes after the first. Once they have claimed a ring, teams may select a Ford Fusion Hybrid car and play a video message from host Phil Keoghan instructing them to make they way to Kanda Myōjin in Tokyo, where they would to find their next clue.
Choice A: Syncing Steps
Teams must make their way to Shinjuku Central Park, where they must learn a series of synchronized "robotic" dance moves performed by members of World Order to the satisfaction of World Order leader Genki Sudo to receive their next clue. If they could not perform the steps properly, they must go to the back of the line before they are allowed to try again.
Aly & Steve, Bergen & Kurt, Hayley & Blair, Jeff & Lyda, Jelani & Jenny, Laura & Tyler, Libby & CJ, Matt & Ashley, and Mike & Rochelle chose this task.
Choice B: Samurai Sake
Teams must make their way to Nakano Sakaue Station Tavern, where they would be shown ten bottles of sake: kanbe, michisakari, momokawa, nanbubijin, okunomatsu, shōchikubai, tōkō, tsukasabotan, yoshinogawa, and zaou. One member must then take an order from a table of samurai, then relay the order to their partner, who must find the correct bottle from a much larger display of bottles. Once they have presented the correct sake bottle to the samurai, they would make a toast before receiving their next clue.
Teams must make their way to Wakaba Higashi Park, where they would find their next clue.
At Wakaba Higashi Park, teams would have the option to U-Turn a team, with two slots available and anonymously. Mike & Rochelle used the first slot of the U-Turn on Jeff & Lyda. Jeff & Lyda attempted to use the second slot of the U-Turn on Jeff & Jackie, however Jeff & Jackie already passed the U-Turn before Jeff & Lyda, thus having no effect on them. Since Jeff & Lyda completed the Syncing Steps Detour, they would be forced to backtrack and complete the Samurai Sake Detour before continuing.
Teams must make their way to the Akihabara UDX Building and search around the vicinity of the building for a marked parking space for the Pit Stop to check in. For coming in first, Jelani & Jenny won the Express Pass.

- The Shinjuku Central Park in Shinjuku was previously visited as the site of the Roadblock in the penultimate leg of The Amazing Race 23.
- The song used during the Syncing Steps Detour option was World Order's titular debut single, "World Order", in which the race utilized their choral dance routine for the task.[4]
- This episode marks the first time in the original American version that a U-Turn has appeared in the first leg of the race.[5]
- Thus, Jeff & Lyda became the first team to get eliminated after a U-Turn on the first leg.
- This episode marks the first time since "Bees Are Much Calmer Than All This!" in The Amazing Race 13 that a Detour is featured on the first leg of the race, as well as the first time that a Roadblock is not featured on the first leg.
- The episode marked the first time in thirteen years that the show was directly preceded by an episode of Survivor, following "My Alarm Clock Didn't Go Off!" in The Amazing Race 2.
