The order of departure from De Koperen Hoogte in Zwolle is as follows:
- Colin & Christie (10:13 pm)
- Tyler & Korey (11:35 pm)
- Leo & Jamal (1:20 am)
- Nicole & Victor (1:55 am)
Teams must fly to London, England, United Kingdom. Upon arrival, they must make their way to the Gatwick Aviation Museum in Charlwood, where they would find their next clue.
Teams must ride a helicopter over the Seven Sisters to Dover Castle, where they would find their next clue.
"Who's can't keep a secret?"
The team member assigned to this Roadblock must decipher a World War II code. They must listen to a speech from former Prime Minister of the United Kingdom Neville Chamberlain and pick out the fifteenth, thirtieth, and fourth words. They must then translate the letters in the code into numbers based on alphabetical order. Under each number was a group of three letters, which they must then rearrange to reveal the final message, type it using a typewriter, then deliver it to a brigadier outside the castle's barracks. If they give him the correct message, "The Dunkirk evacuation has begun. We will never surrender.", they would receive their next clue.

Teams must ride their helicopter back to London. Once there, they must make their way to Masthouse Terrace Pier, where they would find their next clue.
Teams must ride a speedboat on the River Thames to Savoy Pier, then make their way on foot to Piccadilly Circus and find the "golden queen", who would give them their next clue.
Choice A: Know
Teams must make their way to the cabmen's shelter at Kensington Road, where they must board a marked taxi. The driver would take them along a marked route, while giving them a lecture of fifteen streets and seven landmarks on the route, which they must memorize as they would not be allowed to write anything down. Once back at the cabmen's shelter, they must recite the route to Dave Cannell, the Master of the Worshipful Company of Hackney Carriage Drivers to receive their next clue. If they fail to recite the route correctly, they must board the taxicab and go on the route again before being allowed to make another attempt.
Leo & Jamal, Nicole & Victor, and Tyler & Korey chose this task.
Choice B: Row
Teams must take a taxi to Hyde Park, where they must must row a double scull through a 200-meter course on The Serpentine in under one minute to receive their next clue.
Teams must make their way to Camden Market. However, before they can check in, they must search the market for eight marked items seen in a display and use the items to recreate the display in front of the mat. Once finished, they can then check in.

The order from departure from Camden Market is as follows:
- Leo & Jamal (3:53 am)
- Tyler & Korey (4:13 am)
- Colin & Christie (4:29 am)
Teams must fly to Detroit, Michigan. Upon arrival, they must make their way to The Spirit of Detroit monument at the Coleman A. Young Municipal Center, where they would find their next clue.
Teams must take a taxi to the Guardian Building, where they would find their next clue.
"Who's good with numbers?"
The team member assigned to this Roadblock must rappel face-first 500 feet down the side of the Guardian Building. On the way down, they must spot a series of numbers on the building's windows that they would need to unlock a bank vault, along with specific turn-by-turn instructions listed in the building's basement, to retrieve their next clue.

Teams must take a taxi to the Fowling Warehouse in Hamtramck, where they must play a game of fowling. They must throw a football towards each other to knock down bowling pins until all 10 on one side are knocked down, to receive their next clue.
Teams must make their way to the Heidelberg Project, where they must search through a field of clue boxes scattered throughout the outdoor art institution for one that contained their next clue.[1]
Teams must make their way to Third Man Records, where they must properly press five two-tone vinyl records using Race colors. Once five records with a distinct line between colors and labels have been made to a record worker's satisfaction, they would receive their next clue.
Teams must take a taxi to Hart Plaza, where they must search among a group of Rockin' 1000 musicians to find their next clue.
Teams must assemble a five-piece drum kit, following a finished example which they were not allowed to touch. As they build, the musicians will play The White Stripes single, "Seven Nation Army". Once their drum kit is correctly assembled, the lead drummer would give them their final clue.
Teams must make their way to Fort Wayne and cross the Finish Line.

- Due to the season premiere of Big Brother 21, this episode has moved back on Wednesday at 9 pm.
- In Leg 11, Leo & Jamal broke Rachel Reilly's record for most legs run in their The Amazing Race career.
- At the end of Leg 11, with Nicole & Victor's elimination, Christie Lee Woods is the last female standing.
- The final task was supposed to take place at Belle Isle Park, but had to be moved after the permit was denied.[2]
- As of the final leg, all remaining Race teams have improved from their previous placement. Coincidentally, all three teams improved by one placement.
